The second vowel sound that substitutes "R" es the cardinal 26. This vowel is long and today I do have a video of the "elegant woman" from the BBC.
Now this vowel is one of the easiest to pronounce, but the difficult thing comes at the time o listen at it. At first it makes no sense to our ears, and we can't find much of a difference between this and the Schwa sound, but believe me, there is not a big difference between those two, just make /ɜ/ longer for now and after some time you will start to notice the difference. You might pronounce the difference since the first time with out noticing.
This vowel usually appears in stressed* syllables:
Murderer /'mɜː.də.ɹə/
Alternative /ɒl'tɜː.nə.tɪv/
University /juː.nɪ'vɜː.sɪ.ti/
So, what happens with /ɜ/in rhotic environments?
/ɜ/ also has a coloured vertion, a rhotic pair wich is /ɝ/.
And what's the difference between coloured /ɝ/ and /ɚ/?
well /ɝ/ will appear in stressed syllables and so it will have more emphasis and length.
Murderer /'mɝ.dɚ.ɚ/
Alternative /ɒl'tɝ.nə.tɪv/
University /juː.nɪ'vɝ.sɪ.ti/
*Remember Stressed syllable is what we call in spanish sílaba tónica.
